Xgene's CeSSE Technology
Xgene’s cell-sorted organ regeneration technology provides a new, simple method for creating a so-called full-thickness skin equivalent. It relies on the body’s natural tissue-building mechanisms to re-create the bi-layer human skin equivalent, AccuSkin, Xgene's proprietary in vitro skin product.  By omitting exogenous collagen gel to encapsulate dermal fibroblasts in our patented cell-sorted organ regeneration technology, the direction and orientation of how the homogenously mixed epithelial and mesenchymal cells migrate relative to each other can be driven by our proprietary system.

 

AccuGraft™

We have created AccuGraft™, a upside-down version of AccuSkin™, where the epidermal cell layers are juxtaposed to the transwell filter, and the dermal cell layers are on top of the epidermis.  AccuGraft™ is suitable assay platform for applications such as wound care in vitro product tests.  This prototype AccuGraft™ model can be served as the basis for future medical skin graft development based on its unique feature allowing possible direct interaction between dermal fibroblasts from AccuGraft™ and tissues from the wound beds.

Standard AccuGraft™ Products

Our standard premiere product AccuGraft™ is a “full-thickness” bi-layer skin assay system composed of the epidermal and dermal layers.  AccuGraft™ has a niche to recapitulate epithelial-mesenchyme cross-talk because this is the only in vitro upside-down skin equivalent platform offered in the market.  Since Xgene's patented cell-sorted organ regeneration technology do not employ any exogenous matrix nor scaffold to encapsulate the fibroblasts, it eliminate the physical difficulty of creating of bi-layer skin system with the dermis side up.

Standard AccuGraft™ plate is currently offered in 96-well formats with 32 wells per plate using normal human primary keratinocytes and fibroblasts as the input cells.  Complimentary maintenance medium and instruction sheet will be provided in each AccuGraft shipment.

Custom AccuGraft™ Services

Custom-tailored AccuGraft™ can be reconstructed with a wide choice of skin cell characteristics. For example, the input cell types can be derived from young vs. old donors, normal vs. disease-afflicted skin biopsies, and/or genetically engineered cells, such as siRNA knockdown cells.  For adaptations to your companies' specific needs, please inquire.

  • Absolutely NO Testing on Animal Skin!  Many of the existing tests done in animals can be safely replaced by in vitro products.  More details can be found at the ECVAM and ICCVAM web sites under useful links.

  • AccuGraft™ can be adapted to high-throughput screening formats and have shown responses on broad classes of compounds similar to human skin.  Possible applications of AccuGraft™ in the following areas to expedite target selection and validation processes during drug development are listed:

    • toxicology

    • pharmacological drug efficacy and responses

    • irritancy

    •  wound care products

    • wound-healing related disease modeling

  • AccuGraft™, with additional development & FDA approval, may also have potential medical grafting applications.
